"Prep1 Homeodomain Transcription Factor: A New Developmentally Essential, Tumor Suppressor that Controls DNA Damage"

The seminar will deal with the functions of the Prep1 homeodomain transcription factor. A Prep1 null mutation in mouse leads to early embryonic lethality prior to gastrulation. Studies in hypomorphic Prep1 mice (Prep1i/i) show that Prep1 is a tumor suppressor gene. Indeed, human tumors show at very high (75%) frequency silencing of the gene. Prep1-deficient mouse cells show chromosomal abnormalities, basal DNA double strand breaks but normal DNA repair. Molecular insights into these phenotypes will be described.
