Automated Liquid Handling
Robotic instrumentation for sample synthesis and characterization
The automated liquid handler at the Center for Functional Nanomaterials combines synthesis, purification, and characterization of materials through coupling an ecosystem of instrumentation with a versatile liquid handler (Tecan Fluent 1080). Workflow on this one-of-kind robot can be customized to the material sets and needs of the user, including user-defined AI-guided and closed-loop experimentation. As a resource for both high-throughput experimentation and workflow or assay standardization, its customized setup allows users to produce and characterize materials in a controlled, consistent, and timely manner. It has three arms – two for liquid handling and the other for the transfer and handling of different classes of labware. Liquid-handler workflows can be connected to additional research instruments and expertise found at CFN and can evolve to incorporate new synthesis and characterization tools, encouraging collaborative research.
Connected Instrumentation
Disposable Tip, Air Flexible Channel Arm (aFCA)
Ideal for the preparation and distribution of samples and controls, the Air FCA's interdependent pipetting channels can access any tube or well configuration, with automatic adjustment of tip-spacing between labware formats. High-definition liquid handling and air displacement technology ensures precision and accuracy over a wide dynamic range, from sub-microliter to milliliter volumes. Patented Adaptive Signal Technology precisely detects even small volumes, enables accurate monitoring of liquid, and allows the use of smaller reagent and sample volumes for dramatic cost savings. Each pipetting channel is protected by an inline filter that prevents liquid being drawn into the channel. The eight independent channels are equipped with disposable tips and a low disposable ejection option to eliminate splashing or aerosol during tip ejection.
Fixed Tip, Liquid Flexible Channel Arm (liq FCA)
Ideal for the preparation and distribution of samples and controls, the FCA's interdependent pipetting channels can access any tube or well configuration, with automatic adjustment of tip-spacing between labware formats. High-definition liquid handling ensures precision and accuracy over a wide dynamic range, from sub-microliter to milliliter volumes. Patented Adaptive Signal Technology precisely detects even small volumes, enables accurate monitoring of liquid, and allows the use of smaller reagent samples volumes as well as viscous solutions.
Robotic Gripper Arm (RGA)
Robotic gripper arm used to pick and transfer objects within the working area of the Fluent. It can be equipped with dedicated gripper fingers for rapidly moves plates between logistical and processing devices without interrupting the pipetting process. It smoothly and quickly delivers labware to pipetting arms and integrated devices.

Plate Centrifuge
Hettich Rotanta 460 Robotic, Refrigerated
- The centrifuge is mounted under the worktable. It can be loaded and unloaded with the robotic gripper arm. Temperature range: +6°C to +40°C.

Tube Centrifuge

- The centrifuge is mounted under the worktable. It can be loaded and unloaded with the robotic gripper arm.

TE-Chrom Segment
Batch elution or fraction collection. Suitable for applications such as bioprocessing, method development, condition screening, resin screening, step gradient elution, sample prep for mass spec, binding studies, sample concentration and PAT. Currently in use with OPUS Robocolumns from Repligen.

Ziath DataPaqTM Express
The Ziath DataPaqTM Express Rack Reader is a 2D datamatrix tube camera reader with an image capture time of only one second. Compatible with all 2D barcoded tube racks in SBS format, the DataPaqTM Express is the smallest rack scanner currently available. Its camera based instrument delivers rapid imaging and decoding.

The unit shakes from 100 to 2000 rpm, with an amplitude of 2 mm, ideal for 96, 384, and 1536 microtiter plates. It has an automatic slow start-up which avoids spillage.

Vial Gripper
The capper/decapper is a device that when commanded will hold a vial in position for capping or uncapping. The grip of the fixture is adjustable to prevent damage to sensitive vials. The fixture is designed to accommodate 15mL and 50mL tubes.
Tecan Spark Plate Reader
Capable of UV/Vis spectroscopy and fluorescence measurements.

BioTek Cytation 5 Imaging Multimode Reader
Currently not directly incorporated into liquid handler but can be included in workflows. Automated digital widefield microscopy with conventional multimode microplate reading.

- The instrument possesses bright-field (4X, 20X, 60X), fluorescence (4X, 20X, 60X), and phase-contrast (20X) imaging

- Capable of laser autofocus to ensure consistent sample focus
- Conventional UV/Vis spectroscopy and fluorescence measurements with modifiable bandpass on excitation and emission. The instrument has a red-shifted PMT capable of spectral collection up to 900 nm in 1 nm increments
- Control plate shaking and temperature to 65°C with a 4-zone temperature control system, with the ability to set plate-lid temperature gradient to prevent condensation on plate lids/seals.

The automated liquid handler is driven by a customized Tecan Fluent 1080. The workbench has three robotic arms, specifically an 8 channel Air Flexible Channel Arm for disposable tip handling, an 8 channel Liquid Flexible Channel Arm with fixed tips for up to 5 mL liquid handling (capable of positive pressure and handling viscous solutions), and a robotic gripper arm with three finger options (eccentric, center, and tube grippers). All tips are capable of liquid level detection.
